Metolose SR — a high viscosity HPMC for retard formulations.

Dr. Jörg Brunemann, Managing director, Syntapharm, Germany

The hydrophilic matrix system is a simple sustained release technology for oral dosage forms, economical and easy to establish. High viscous HPMC is widespreadly used for the development of such matrix systems.

A key point for the success is the homogeneous and uniform distribution of the particles in the system. The reproducibility of the drug release is influenced mainly by the HPMC properties, especially the substitution level, the particle size distribution and the viscosity.

Metolose SR is exclusively designed for the preparation of hydrophilic matrix systems with tight specifications for the most critical parameters.

The presentation is divided in three parts.
First some general information about hydrophilic matrix systems will be presented. Based on acetaminophen as model drug, the influence of certain HPMC properties will later on be demonstrated. Finally a comparison between direct compression and wet granulation as possible production techniques for matrix tablets using Metolose SR will be given.
