Polyplasdone — not only a simple desintegrant.

In daily life formulators would use excipients that are used as a standard in their laboratories. This might be often a good decision but not always. Choosing the proper disintegrant for instance during the development of tablet formulations might have strong impact on the characteristics of the tablets being formulated. Factors such as physical characteristics of the disintegrant and their impact on mechanical characteristics of the tablet, interaction with the disintegration medium, possible entrapping of the drug itself should be taken seriously into consideration, especially when high amounts have to be used as for example in the formulation of Orally Disintegrating Tablets. Different disintegrants utilizes different disintegration mechanisims .Polyplasdone® (crospovidone) combines swelling and wicking (porosity and capillary action) mechanisms which contributes to its high swelling and wiching necessary for effective disintegration.

Polyplasdone has unique characteristics which makes it formulators user friendly excipient because of the following properties: porous particle morphology , high density cross linking which prevents gelling when fully hydrated , availability in two particle size (XL & XL-10) grades which provides the formulator with flexibility to choose the optimal grade for best performance , brings excellent disintegration performance to wet and dry granulations and direct compression formulations, improves tablet stability, hardness and friability and reduces capping, possesses high compressibility , does not form ionic complexes with ionic drug actives that may retard drug release, disintegration is not impacted by changes in pH. In orally dispersible tablets ( ODT) formulations which requires high percentage of disintegrant polyplasdone XL-10 grade gives good compression, low friability and smoother mouth feel .

Case studies are going to be presented highlighting these different aspects and demonstrating that the selection of the suitable disintegrant would help the formulator achieve best results. Polyplasdone® (crospovidone) grades are the excipients of choice in many cases, and this is not only for being simple disintegrants..
